Abstract:
We describe a novel method for the isolation of human erythrocyte membranes based on the covalent binding of the cells to a solid support via cleavable disulfide bonds. After attachment, the cells are lysed, washed and the resultant membranes detached from the support by mild reduction with dithiothreitol. The application of this method to the isolation and identification of outer cell surface proteins is described.
