Abstract:
Intracellular nucleotides of the achlorotic flagellate Astasia longa were fractionated by anion-exchange chromatography from cultures grown logarithmically at 14.5 ° and 28.5 °C, the two temperatures used for the synchronization of cell division of the protozoa, and also from synchronized cultures after the burst of division. No large changes in the relative quantities of the various nucleotides were observed under these conditions, but the total pool size increased markedly after the burst of division. Thirty-four nucleotides were obtained in sufficient quantity to be examined, and twelve of these were identified. These include two possible precursors of NAD, thereby suggesting the pathway of biosynthesis of NAD in Astasia. Some properties of the other twenty-two nucleotide components were established by spectral analysis and by paper chromatography, and some were tentatively identified. © 1966.
