Metaphase chromosomes isolated from an inducible SV40 transformed Chinese hamster cell line with no detectable infectious virus or free virus DNA, were transferred into SV40 permissive BSC1 cells. This chromosome transfer resulted in the appearance of SV40 T-antigen in one per 104 treated cells and in the activation of infectious virus in one per 4 × 105 treated cells. No appearance of T-antigen or virus activation was obtained with sonicated chromosomes, whole nuclei, DNA, or lysates from the transformed cells. The results show the chromosomal location of the SV40 genome and the activation of viral genes after chromosome transfer. © 1974.
