Abstract:
DNA was extracted from single soil-borne seeds of Orobanche ramosa L., O. crenata Forsk, O. cumana Loeffl, and O. aegyptiaca. Species identification was then possible by RAPD-PCR, based on specific DNA markers, and could be verified by Southern hybridization with specific probes. Seed identification was also possible for dead (tetrazolium-negative) seeds. This method is a powerful diagnostic tool for Orobanche classification in agricultural fields.
