The effects of cardiac glycosides, a group of synthetic azasteroids, and erythropheleum alkaloids upon a transport ATPase activity of ciliary processes of rabbit eyes have been studied. All three groups of compounds effectively inhibited the metal-stimulated ATPase activity. The physiological, biochemical, and general chemical similarities of these three classes of compounds make it possible, with careful reservation, to group them together for the adduction of the structural requirements for this pattern of inhibition of the ciliary process enzyme. Quindonium, quinolizinium, and methyl quinolizinium were quite similar to ouabain in their effects. The B isomer quindonium inhibited to a lesser degree while the erythrophleum alkaloids, though highly effective, were insensitive to preparative methods which affected membrane integrity. These results have been discussed with respect to structure-activity relationships, the mechanism of aqueous humor ecretion, and possible use of the azasteroids as ophthalmic drugs. © 1967.
