Abstract:

Growth of unselected, habituated nucellar callus of Shamouti orange (C. sinensis) ceased almost completely on media containing 5 sd 10-5 M 2,4-dichlorophenoxyacetic acid (2,4-D). Six culture lines, two of them originating from gamma-irradiated and four from non-irradiated callus, were selected. Three of the lines grew well up to 2 · 10-4 M 2,4-D, and two of these supported 4 · 10-4 2,4-D in the medium. Stability of tolerance was demonstrated. One line, R-1, has become 2,4-D dependent, while retaining embryogenic capacity in another line was accompanied by cytological change (from diploidy to tetraploidy). Embryos from the selected R-1 line tolerated media containing up to 5 · 10-5 M 2,4-D.
