Abstract:
The gladiolus bud liquid cultures of strain "Dixieland" were studied in modified conical bubble type bioreactor at batch, closed and underfeed culture regimes. The potential of bud culture to produce cormlets from apical domes was expressed by coefficient of specific cell mass (qa, mg/cormlet) calculated as ratio of initial organ culture biomass on the stage of bud regeneration, to the total number of cormlets which were obtained from this mass. Based on the experimentally determined qa values, the parameter of cultivation process in bioreactor-cormlet productivity (P c, cormlet/1 day)-were estimated as ratio of biomass productivity (P, g FW/1 day) to qa-At the closed culture regime in bioreactor, when the initial concentration of biomass was ∼140 g FW/1 and the final one ∼420 g FW/1 in stationary phase Pc was ∼6800 cormlet/1 day. It was shown that the number of apical domes was proportional to the addition of the gladiolus bud culture biomass at batch and closed regime, and could be increased at underfeed culture when had no addition of the gladiolus bud culture biomass.
