Abstract:
Radiolabeled polar ecdysteroids were isolated from Manduca sexta pupae and applied to a column of the ion-exchange adsorbent DEAE-Sephadex. Elution with a NaCl gradient solvent system efficiently separated the polar ecdysteroids into three groups, eluted in order of their increasing ionic strength: free ecdysteroids, ecdysteroid acids, and ecdysteroid conjugates. The separation method afforded excellent sample recovery and purification, and facilitated HPLC analysis of the ecdysteroid acids which may be key inactivation products of molting hormones in M. sexta.
