Abstract:
In the framework of a study aiming at producing transgenic grapevines resistant to viruses of the rugose wood disease complex, the movement protein (MP) genes of Grapevine virus A (GVA) and B (GVB) were analysed in transgenic Nicotiana plants. The cassettes transcribing MP genes in their sense (MP+) and antisense (MP-) orientation were transferred to Nicotiana benthamiana and N. occidentalis by Agrobacterium tumefaciens-medmted transformation. Regenerants expressed the RNA transcripts. Rl seedlings expressing the antisense gene, when challenge-inoculated with the homologous virus showed a consistent reduction of virus titer, and did not show symptoms up to 18 days post inoculation. MP+ lines showed lower ELISA readings and percentage of symptomatic plants as compared with the non-transgenic control. Both constructs were successfully inserted in somatic embryos of Vitis rupestris and V. vinifera cv. Superior seedless, from which several plants were regenerated.
