Abstract:
Proteolytic activity of centrifuged sap extracted from fresh maize leaves was studied by its action on the synthetic substrate carbobenzoxy-l-tyrosine p-nitrophenyl ester. The crude sap preparation was fractionated on a DEAE-cellulose column using an ascending gradient of NaCl concentrations in phosphate buffer pH 6·2. The eluted peak in esterolytic activity corresponded to that of the proteolytic action on casein, as well as to a peak in protein concentration. Similar results were obtained with sap of tobacco leaves. © 1965.
