Abstract:
A simple procedure for the preparation and purification of trans-[14C]geranylgeranyl pyrophosphate from dl-[14C]mevalonate is described. This procedure involves biosynthesis of trans-[14C]geranylgeraniol from dl-[14C]mevalonate by cell-free extracts of the fungus Gibberella fujikuroi and chemical synthesis of trans-[14C]geranylgeranyl pyrophosphate from trans-[14C]geranylgeraniol. The purification of trans-[14C]geranylgeraniol is achieved by chromatography on a silver nitrate-impregnated silica gel column. The pyrophosphate ester is purified on a silica gel column. © 1979.
